Introduction

Fill each of the PTM tanks with 1.5 L of the appropriate buffer (H or L).

[Optional, use if you wish to program a preheating] In the menu “1 – Set Clock and Delay Start” set the current day and time.

In the menu “2 – Setup Cycle (Time and Temp)” set the “Time to run” to 01:00 h and the “Temperature” to 102 °C, or to the optimal time and temperature for your tissue and markers.

In the tab “3 - Preheat Settings”, set the “Preheat Temp” to 85 °C and the “Cool End Temp” to 65 °C.

[Optional, use if you wish to program a preheating] Set the desired “Preheat Start Time” and select the preheat mode “One Time” or “Continuous” for a single or daily preheating.

Place the lid on the tank, latch the PTM lid.

If the PTM was not preheated previously, press “Run” in the home tab to start the preheating of the tank(s)

Once 85 °C is reached, mount the slides into the slide rack. Open the PTM, remove the tank lid and insert the slide rack. Place lid back and latch the PTM lid.

Press “Run” in the home tab to start the Heating cycle.

When the heating cycle is finished and the tank has cooled down to 65 °C, a beep will sound, and the lid will unlock: take the rack(s) out of PTM and rinse the slide(s) with Lunaphore Multistaining Buffer.

Place and keep the slide(s) in fresh Multistaining Buffer at room temperature until staining.
