Introduction

This guide from the Center for Microscopy and Image Analysis, UZH, Zurich, Switzerland shows how to set up Atlas acquisition with EPU at the transmission electron microscope Thermo Titan Krios G31.

  1. Start an inventory, clicking "inventory" in the Autoloader Tab.

    • The Inventory will see which slots in the cassette are occupied

    • Empty slots are shown in grey.

    • Full slots are shown in blue.

  2. Import you EPU presets. Set The Atlas presets to the microscope. Load the first grid on the campus stage, selecting the grid slot and pushing load.

    • If you don't load the first grid yourself, everything will crash!!!

    • Open the column valve.

  3. Select the Atlas tab > "Session Setup" task. In the Ribbon Bar, select New Session. Select a name for you session.

  4. Select the Atlas > "Screening" task. Example of occupied Slot.  You can rename the occupied slots with your sample name. Example of Empty slot.

  5. Select the slots you want to include in the screening session by clicking on the little square close to the slot number. Selected grids would show a tick ("v"). To speed up the screening procedure, in the autoloader tab in the microscope computer, set the "Turbo Always On" option. Start the screening.

  6. If the the current grid looks really bad halfway through the Atlas and you wish to skip it (but continuing acquiring the Atlases of the next grids), you can do so with the skip option. If you wish to stop the screening session, select the stop option.

    • Completed Atlas acquisition for the slot will be shown with a green panel. To load and observe the Atlas, double click on the slot.

  7. Select your favorite grid and load it on stage, with the "Load Sample" option. It is important to load your grid with EPU and not from the autoloader interface.

    • If you load the grid with EPU, the program will correct for the movement caused by loading and unloading for the campus stage. If you instead load the grid from the autoloader tool, there would be an offset between the positions in the Atlas and the actual positions.

    • Put back the "Turbo Auto Off", in the autoloader option, so that you won't have vibrations during data collection.
