Start heating at least 30 min before you start your experiment. This allows the chamber and the microscope components to heat up and avoids major drift during your experiment.

Locate CO2 gas bottle close to the door

Open the main valve

Open the small valve

Locate the tubing in between Leica tables and open the blue valve for Stellaris.

Check water level in humidifier

If the water level is at minimum refill it from the stock bottle on the shelve above the microscope

Switch on the touchscreen of the okolab

Check the desired temperature in the box

If you need to change it. Click on the temperature and adjust +/-

Confirm Set

Once the temperature/level of CO2 is stabilized the yellow dot will turn green. For that you have to make sure, that environmental box is fully closed.

Find a live-cell chamber in the tower next to the microscope.

You can remove the magnetic lid and connect the tubing to the chamber.

Install the chamber on the microscpe

It is important that this tubing will around the galvo stage, otherwise it won't sits perfectly in the stage.

Block the other openings for dish/microscopic slide, if you don't use them. Otherwise the CO2 will escape from the chamber

Put the lid on top of the chamber

Close the environmental box around microscope to let temperature stabilize
